Major equipment and facilities such as high performance liquid chromatography, (HPLC - Waters), Gas chromatography (G.C.), P.C.R., Lab scale Fermenter, Uv-vis-Spectrophotometer, D.N.A. & protein Electrophoresis system, high speed refrigerated centrifuge, Light microscope with projection system and (F.T.I.R.S & A AS - under acquisition)


Advanced Techniques Training

Advanced Techniques Training
Six Weeks - (Fee Rs. 8000/- per Student)

INSTRUMENTATION UV/ visible spectrophotometer, PCR, Electrophoresis transilluminator, laminar air flow, microscope, pH meter, autoclave, centrifuge, shaker, water purification system, auto pipettes, weighing balance etc.
  • Basic knowledge of designing, performing and analyzing the experiments
  • Collection of samples from different sources using different techniques
  • Media preparation and sterilization techniques
  • Studying the type of microbial flora of different samples
  • Staining techniques (simple staining, differential staining, Gram staining, acid fast staining.
  • Screening of organisms for the production of enzyme producing isolates (Proteases, Cellulases, Xylanases, Amylases, Lipases, Mananases etc).
  • Shake flask fermentation for enzyme production.
  • Identification of isolates by biochemical methods.
  • Monitoring the growth and enzyme production.
  • Enzyme production at lab scale fermenter
  • Basic characterization of the enzyme.
  • Isolation of plasmid DNA
  • Isolation of genomic DNA
  • Agrose gel electrophorese.
  • Testing the pharma products eg Tablets, Capsules, Syrups, injections etc.
  • Introduction and principle of HPLC and its various components
  • Introduction to Gas Chromatography (GC).

INSTRUMENTATION UV/visible spectrophotometer, laminar air flow, microscope, pH meter, autoclave, centrifuges, shakers, water purification system, auto pipettes, weighing balances, Electrophoresis apparatus, Transilluminater, PCR etc.
  • Introduction to recombinant DNA tools and genetic engineering techniques
  • General rules to perform molecular biology experiments
  • Preparation of solutions for the molecular biology experiments.
  • Isolation of plasmid, genomic, metagenomic DNA.
  • Agrose gel electrophoresis of isolated DNA.
  • Spectrophotometeric estimation of quantity and purity of isolated DNA.
  • Restriction digestion of DNA.
  • Ligation of fragmented DNA in the plasmid.
  • Trasformation and screening of recombinants
  • Selection of the recombinant of interest by masterand replica plating.

INSTRUMENTATION UV/ visible spectrophotometer, laminar air flow, microscope, pH meter, auto claves, centrifuges, shakers, water purification system, auto pipettes, weighing balances etc.

  • Preparation of microbiological media (Basal medium, enrichment medium, selectionmedium etc.)
  • Sterilization techniques (Dry heat sterilization, moist heat sterilization, chemical sterilization).
  • Culture techniques (Spread plating, pour plating, streaking, stab cultures).
  • Shake flask fermentation for enzyme production.
  • Staining techniques (simple staining, differential staining, Gram staining, acid fast staining.
  • Wet mount and hanging drop preparation.
  • Identification of bacterial isolates by biochemical methods.
  • Preservation of microorganism .
  • Processing the different clinical samples.
  • Antibiotic sensitivity testing of different clinical samples.
  • Microbiological examination of water

(A)INTRODUCTION & PRINCIPLES of HPLC - G.C. AND THEIR COMPONENTS.
  • The various Modes of chromatographic separation(Reversed phase, Normal phase etc.)
  • Understanding working of HPLC, degassing the mobile phase and preparation of sample.
  • To control retention and selectivity in reversed-phase LC.Concept of gradient elution
  • Analytical method validation and troubleshooting
  • Introduction to gas chromatography(GC). Column types and selection. Sample introduction techniques
  • Reviewing , liquid, and other injection methods. FID detection systems
  • Analysis and interpretation of results. Routine maintenance and troubleshooting

(B)INTRODUCTION TO FERMENTER FUNCTIONING
  • Media formulation for enzyme production
  • Sterilization of fermenter, media and ancillary equipments
  • Demonstration of Fermenter functioning
  • Inoculation of media
  • Monitoring the growth &Enzyme production
  • Decontamination of fermenter & ancillary equipment

INSTRUMENTATION HPLC, GC, UV-VIS spectrophotometer, dissolution app., pH meter, auto pipettes, weighing balance, centrifuge, water purification system etc.
  • Testing of pharma products eg. Tablets, capsules. Syrups, injections etc.
  • Wet chemical analysis of Pharma Products.
  • Qualitative/ Quantitative analysis of chemical sample
  • Practicals in Volumetric/ Gravimetric/ Colorimetric analysis Water analysis

  • MBR (Methylene Blue Reduction test)

  • SPC (Standard plate count)
  • DMC (direct microbial count)
  • Rapid plate form tests
  • To determine the bacteriological quality of various food products eg. Ice-cream, butter, condensed milk, etc.
  • Quantification of reducing, non reducing, and total sugar in various foods eg. Milk & milk products, fruit juice, etc.
  • Quantification of starch in various food products eg. Cereals.
  • Quantification of fat content in various food products.
  • Introductions to Fermentation and system types, Media formulation for enzyme production.
